GlnB is the archetypal PII protein; a ubiquitous family of small trimeric signal transduction proteins that facilitate regulation of bacterial nitrogen metabolism by controlling the activities of a variety of enzymes, transcription factors and membrane proteins. In nitrogen limitation GlnB is subject to post-translational modification by uridylylation (mediated by GlnD) of Tyr51 in the T-loop of each subunit. This modification is reversed in N-sufficient conditions. GlnB interacts with and regulates the activities of the histidine protein kinase NtrB, which controls phosphorylation of NtrC. It also regulates the activity of adenylyltransferase (GlnE) which in turn regulates the activity of glutamine synthetase (GlnA). glnK is a glnB paralog.
