E. coli K-12 wildtype is derived from a strain in the collection of the what was then the Department of Bacteriology at Stanford University. In 1944, Gray and Tatum[1] reported the isolation of a biotin auxotroph and a threonine auxotroph after X-ray mutagenesis of E. coli K-12. The parental strain is described in the acknowledgements as having been obtained from Dr. C.E.Clifton at Stanford. Elsewhere, Tatum and Lederberg describe the strain as having been used in student labs at Stanford for several years[2].

The original K-12 strain was F+ and lysogenic for phage lambda.

Different stocks of the original K-12 strain were obtained by different labs at different times. The Coli Genetics Stock Center has two isolates of E. coli K-12 wildtype: WG1 and EMG2. WG1 was obtained from J. Lederberg and EMG2 was obtained from Clowes and Hayes. The stock center isolate of EMG2 is described as having no mutations, while the isolate of WG1 available from CGSC has an rfbB deletion and carries F1-1, a shortened variant of F.
