The expression vector pBS(Kan) was constructed by removing the kanamycin resistance cartridge from pKG1022[1] by digestion with KpnI and BamHI, blunt-ending the fragments using T4 DNA polymerase, and then blunt-ligating into the ScaI site in the middle of the ampicillin resistance gene of pBluescript II KS- (Stratagene, La Jolla, Calif.).[2]
