Construction

Most of the deletions in MDS42 were constructed by lambda Red-mediated recombination of linear fragments into the genome, followed by removal of exogenous sequences by double strand break (DSB)-stimulated recombination.[1] The individual deletions are described in the Supporting Online Material.

Notes

Compared to MG1655, MDS42 has 663,316 bp deleted (14.3% of the original genome).[1] Deletion of fhuACDB confers resistance to phage T1. The endA gene was deleted to improve the integrity of plasmid DNA isolated from this strain.
