Notes
YA027 was constructed from K-12 MG1655 (CGSC #6300). YA027 has a 524-kbp-long partial duplication of the chromosomal region between IS2F (yi22_6) and IS2A (yi22_1). One copy of the duplicated region has the PaeR7I RM genes and a linked KmR gene inserted into lacZ, while the other copy of the duplication has a CmR gene inserted into lacZ. See the figure below, which is based on Figure 1A of Asakura et al. (2011).
YA027 was constructed by P1 transduction. "The recipient strain, YA001, carried PaeR7I RM genes together with the Kmr gene inserted into the EcoRV site in the lacZ gene (1) of MG1655, and the donor strain, BIK2158, carried the Cmr gene inserted into the same EcoRV site in the lacZ gene. After transduction, clones simultaneously Kmr and Cmr were selected and one was designated YA027."
