Notes
The pTXB1 is an E. coli vector created for expression of recombinant proteins and subsequent purification. A target gene can be inserted into the the ORF by use of the MCS upstream of the Mxe intein. The chitin-binding domain(CBD) downstream of the Mxe intein results in a target protein-intein with C-terminal chitin beads that facilitate protein purification. Release of the protein is induced by the thiol cleavage activity of the intein.
History
Derived from pBR322, pTXB1 possesses the Mycobacterium xenopi GyrA intein; with 198 amino acids, the Mxe Gyr A gene is the smallest of any known elements for active protein splicing that is found to occur naturally.
