The plasmid that contains that pGL2-Promoter Vector also contains a luciferase gene. The pGL-2 Promoter is located upstream from this gene. In the promoter-luc transcriptional unit DNA fragments can be inserted. These contain putative enhancer elements which can be inserted upstream or downstream from the promoter-luc unit.
The pGL2-Promoter along with other pGL2 luciferase reporter vectors are factors that could potentially regulate mammalian gene expression. These vectors provide a basis for quantitative analysis for these contributing factors. These factors include cis-acting or trans-acting. An example of the two factors would be promoters and enhancers for cis-acting and various DNA-binding factors for the trans-acting factors. Using the the firefly (Photinus pyralis) luciferase contained in the pGL2 vectors the transciptional activity in transfected cells can be monitored. The pGL2 vectors also contain features that are able to aid in the characterization and mutagenisis of the putative regulatory sequences.
