pGEM-5Zf(+) and pGEM-5Zf(–) contain filamentous phage f1 origin of replication. These vectors can be used for standard cloning, circular ssDNA production, and as templates for in vitro transcription. Within the α-peptide coding region of β-galactosidase is included a multiple cloning site enclosed by T7 and SP6 RNA polymerase promoters on either side. Inactivation of the α-peptide through insertion allows for the simple blue/white color screening for recombinant clones. The multiple cloning region arrangement is designed for generating unidirectional deletions with Promega's Erase-a-Base® System. pGEM-5Zf(+) and pGEM-5Zf(-) differ in the f1 origin orientation.
