The pG5luc vector is part of a two-hybrid system. This two-hybrid system in particular is called the checkmate mammalian two-hybrid system and is extremely powerful when detecting protein:protein interactions in vivo. The main feature of the hybrid system is the modular domains found in some transcription factors. These domains are the DNA-binding domain and a transcriptional activation domain. The DNA-binding domain binds to specific DNA sequences and the transciptional activation domain interacts with the basal transcriptional machinery. RNA polymerase II complexes assembly is promoted at the TATA box and increases transcription when a transcriptional activation domain is in association with a DNA-binding domain. The Checkmate Mammalian two-hybrid system recieves the DNA-binding domain and the transcriptional activation domain from different and separate plasmids. They are closely associated when one protein fused to a DNA-binding domain interacts with a second protein fused to a transcriptional activation domain. The interaction between the two proteins results in transcription of a reporter gene.
