Notes

Each pET200/D/LacZ plasmid carries a T7/lac promoter which provides the plasmids with a promotor for high-level expression. The lac operator sequence is located immediately downstream from the T7/lac promoter. The gives the cell added regulation of basal expression. The origin is that of the low-copy pBR322 of replication and is contained in all vectors. In the uniduced state the origin reduces the basal expression level. Also, each vecotr offers flexible options for simplifying protein detection, cleaving purification tags, selecting plasmid-carrying clones, and improving protein yields.

History

There are five plasmids that carry the same features but vary very slightly between them. These are pET100/D-TOPO®, pET101/D-TOPO®, pET102/D-TOPO®, pET151/D-TOPO®, and pET200/D-TOPO.
