Notes
The Champion pET-DEST42 Gateway destination vector contains a strong T7 promoter that controls the high-level prokaryotic expression. The vector is designed for rapid cloning in conjuction with a Gateway entry clone which uses a lamba phage site-specifc recombination. The T7 RNA polymerase initiates expression in BL21 E. coli. There are several specific functions offers by the Champion pET-DEST42 including the bacteriophage T7 promoter for high-level expression, and O operator sequences for lac repressor binding, pBR322 origin for minimal basal expression, C-terminal V5 and 6xHIS tag for efficient detection and purification, and R sites for efficeint recombination with any attL-flankied Gateway entry vector. The use of the O operator sequence for lac repressor binding makes sure that the regulation is very tight for transcription.
