It is now known that superinfection at high frequency can occur with toxigenic lambda-like phages, and here we demonstrate that superinfection of a lambda lysogen can lead to acquisition of additional lambda genomes, confirmed by Southern hybridisation and quantitative PCR. As many as 8 integration events were observed, but at very low frequency (6.4 x 10(-4)) and always as multiple insertions at the established primary integration site in E. coli. Sequence analysis of the complete immunity region demonstrated that these multiply infected lysogens were not immunity mutants. In conclusion, although lambda superinfection immunity can be confounded, it is a rare event.</protect>
==Significance {{HelpIcon|Help:Literature}} ==
{{LitSignificance}}
This paper is significant to the study of bacteriophage lambda and closely related phages because it demonstrates that immunity to superinfection in lambda is not absolute <ref>Mark Ptashne. A genetic switch. Cold Spring Harbor, N.Y.: Cold Spring Harbor Laboratory Press, 2004. isbn:0879697164.</ref>, though the mechanism for this is likely not via integrase mediated site specific recombination but rather homologous recombination within the resident prophage. This is in stark contrast to other bacteriophages deemed to be lambdoid (Stx-phages), which can actively promote integration of superinfecting phages into distinct sites throughout the genome at high frequencies <ref name='PMID:18048923'/> <ref name='PMID:12761125'/> <ref name='PMID:18469095 '/>. Therefore, the assumption that all lambdoid phages must exactly abide by the canonical rules, based on lambda itself, is unfounded.
This paper is significant to the study of bacteriophage lambda and closely related phages because it demonstrates that immunity to superinfection in lambda is not absolute <ref>'Ptashne-Genetic-Switch isbn=0879697164'</ref>, though the mechanism for this is likely not via integrase mediated site specific recombination but rather homologous recombination within the resident prophage. This is in stark contrast to other bacteriophages deemed to be lambdoid, which may actively promote integration of superinfecting phages into distinct sites throughout the genome at apparantly high frequencies <ref name='PMID:18048923'/> <ref name='PMID:12761125'/> <ref name='PMID:18469095 '/>. Therefore, the assumption that all lambdoid phages must exactly abide by the canonical rules, based on lambda itself, is unfounded.
|{{TableHeadingColor}} |'''Abstract''' || <protect>During the course of experiments aimed at identifying genes with ribonuclease III (RNase III)-dependent expression in Escherichia coli, we found that steady state levels of bdm mRNA were dependent on cellular concentrations of RNase III. The half-lives of adventitiously overexpressed bdm mRNA and the activities of a transcriptional bdm'-'cat fusion were observed to be dependent on cellular concentrations of RNase III, indicating the existence of cis-acting elements in bdm mRNA responsive to RNase III. In vitro and in vivo cleavage analyses of bdm mRNA identified two RNase III cleavage motifs, one in the 5'-untranslated region and the other in the coding region of bdm mRNA, and indicated that RNase III cleavages in the coding region constitute a rate-determining step for bdm mRNA degradation. We also discovered that downregulation of the ribonucleolytic activity of RNase III is required for the sustained elevation of RcsB-induced bdm mRNA levels during osmotic stress and that cells overexpressing bdm form biofilms more efficiently. These findings indicate that the Rcs signalling system has an additional regulatory pathway that functions to modulate bdm expression and consequently, adapt E. coli cells to osmotic stress.</protect>
